Calcein staining of calcified structures in pearl oyster Pinctada margaritifera and the effect of food resource level on shell growth ArchiMer

Marine mollusc shell growth has been widely measured using fluorochrome marking. In order to test the efficiency and reliability of calcein staining on P. margaritifera shells and pearls, the present study examined two administration methods, different concentrations and several immersion times.Immersion in a 150 mg.L-1 calcein solution for 12 h to 24 h appeared to be the best method for marking P. margaritifera shells. For pearl marking, injection of a 200 mg.L-1 calcein solution into the pearl pouch was the optimal method. Dans le cadre du programme Reproduction de l’huître perlière Pinctada margaritifera, dont l’objectif est la maîtrise des croisements, le conditionnement en laboratoire des reproducteurs est une étape nécessaire pour espérer contrôler et synchroniser la maturation de ces reproducteurs. Les facteurs présumés qui permettront de contrôler la maturation sont d’ordre environnemental : l’alimentation et la température. Cependant avant d’espérer établir la relation entre la fonction physiologique et les facteurs qui contrôlent cette fonction, il est indispensable de mettre en œuvre une zootechnie adaptée aux besoins vitaux de façon à assurer la survie et une bonne croissance. The pearl industry in French Polynesia is based on exploitation of natural stocks of the black-lip pearl oyster (Pinctada margaritifera); it generates an annual turnover of 90 million euros. Improvements in pearl quality need genetic studies to improve the populations. This pearl oyster is a protandric species, where the sex-ratio normally is biased towards males. There is an increasing interest in gender control to find the mechanisms to augment female proportions for management purposes. Evaluation des conséquences d'une situation hypoxique simulée sur la Capacité Osmorégulatrice des crevettes d'élevage, Penaeus stylirostris et Penaeus vannamei ArchiMer

Les résultats exposés dans ce rapport apportent des informations sur la Capacité Osmorégulatrice des crevettes d'élevage, Penaeus stylirostris et P. vannamei, face à un abaissement de l'oxygénation ambiante. Ces résultats s'appliquent à des juvéniles placés dans des conditions optimum en laboratoire mais à niveau métabolique bas : animaux à jeun, en stade d' intermue C, adaptés après dissipation du stress induit par la pêche et le transfert dans un nouvel environnement. The possible consequences of acidification on pearl farming are disruption of oyster metabolism and change in growth. In the laboratory, we studied the impact of pCO2 (3540, 1338 and 541μatm) on the physiology of pearl oysters exposed for 100 days. This experiment was repeated after an interval of one year. The objective of this study was to observe the impact of temperature on pearl formation using  an integrative approach describing the rotation of the pearls, the rate of nacre deposition, the  thickness of the aragonite tablets and the biomineralizing potential of the pearl sac tissue  though the expression level of some key genes. Fifty pearl oysters were grafted with  magnetized nuclei to allow the rotation of the pearls to be described. Low energy cost for cultured pearl formation in grafted chimeric Pinctada margaritifera ArchiMer

The pearl oyster is one of the rare animal models that support two distinct genomes,  through the surgical graft process operated for culture pearl production. This grafted  organism is assimilated to a chimera whose physiological functioning remains poorly known.  The question of the energy expenditure comparison between chimera and non-chimera  animals arises. To answer this question, grafted and non-grafted pearl oysters were  evaluated for their energetic needs by the indirect calorimetry method. Phenome of pearl quality traits in the mollusc transplant model Pinctada margaritifera ArchiMer

This study was designed to measure responses of four-year-old black-lip pearl oysters (Pinctada margaritifera) to different temperatures and food concentrations and to identify the energy requirements of each sex. Oysters were fed a monospecific microalgal diet of Isochrysis affinis galbana (T-iso). Measurements of oxygen consumption and ingestion rates were carried out at 24 degrees C and 27 degrees C and at two algal (T-iso) concentrations: 5000 and 30 000 cell ml(-1). Response of the blue shrimp Litopenaeus stylirostris to temperature decrease and hypoxia in relation to molt stage ArchiMer

The effect of different levels of dissolved oxygen (DO) and a decreased temperature combined with severe hypoxia were studied in the blue shrimp Litopenaeus stylirostris in relation to molt stage. In the first experiment, shrimps were submitted to DO concentrations of 5 to 1 mg l(-1). Osmoregulatory capacity (OC) was measured in shrimps at molt stage C after 6, 24, or 48 h of exposure. No mortality was recorded but a significant negative effect of DO concentration on hypo-OC was observed for DO concentrations below 3 mg l(-1). Osmoregulation improved after 48 h exposure, compared to 6 and 24 h exposure. In a second experiment, temperature was decreased from 28 to 22 degreesC in 24 h, before a severe hypoxia of 0.5+/-0.1 mg O-2 l(-1) was applied....
